If you've not ridden it before, it's very easy to miss the entrance to the Great River Trail near Marshland. When southbound on Hwy 35 you will see a rest area on the East side of the road up against the bluff. Opposite the rest area is a large wetland that I think has a sign labeling it as Trempealau Preserve. While traveling past this wetland just after the very swampy portion runs it's course is a dirt road on the right. It's not labeled as the Great River Trail in any way by the road. Turn down the road and you will see Great River Trail markers in a 1/2 mile or so. It's also easy to miss the most directly south connection to the trail from this road as well. There will be signs that look like you will need to ride against a One Way guidance, that's what you need to do. That said, if you miss it or want to stop and smell the roses the Perserve has a great overlook over the backwater with a wood dock that extends out and you will see Egrets, Herons, and Eagles much of the time. Missing/skipping the turn will cost you a mile or so. If the Nature Center is open I think you can get your Wis. trail pass there, otherwise your next best opportunity is Perrot State Park. I'd recommend buying one since you will be on the system for days, just in case you run into a Ranger in a non-forgiving mood.

Have fun, looks like a great trip.
